The defending champions had made a shaky start to proceedings, slipping to 41 for three.
But they recovered considerably thereafter, with Lyth (111) and Bairstow (107 not out) combining for a fourth-wicket stand of 205.
At Lord's, Middlesex took control against Warwickshire, thanks chiefly to Sam Robson's unbeaten 175.
There was also 68 from Nick Gubbins - who shared a 180-run first-wicket stand with Robson - to help the home side to 317 for four.
Meanwhile, at Old Trafford, Lancashire debutant Neil Wagner starred as the hosts bowled out Nottinghamshire for 242.
James Anderson could not contribute any wickets, but New Zealand international Wagner took six for 66 to torment the visitors, for whom captain Chris Read (52) and Stuart Broad (43) stabilised things for a while after the score had reached 98 for six.
Lancashire's reply was at 25 for one at close of play.
