Alcacer made his debut for Spain in 2014, going onto score six goals in 13 games since for his country.
The striker also netted 43 goals for Valencia in all competitions in the past two seasons.
The 23-year-old is the first forward to sign for Luis Enrique's side this summer, with the club having also bought defenders Samuel Umtiti and Lucas Digne, midfielders André Gomes and Denis Suárez, and goalkeeper Jasper Cillessen.
Barca's priority was to add a striker but they lost out on signing Nolito and Kevin Gameiro, with the former joining Manchester City and the latter Atletico Madrid.
Alcacer is Barca's fourth attacking option behind Lionel Messi, Luis Suarez and Neymar.
Meanwhile, 20-year-old Munir has joined Valencia for the remainder of the season, with the option to buy the player for 12 million euros at the end of the campaign.
