Clarke, who started his career at Huddersfield, has the option of a further year and becomes Bradford's sixth summer signing.
The 31-year-old is expected to feature in Tuesday night's friendly at Whitby Town.
Former Orient skipper Clarke left Brisbane Road last week having made 131 appearances for the Londoners since signing from first club Huddersfield in July 2012.
He has also had loan spells at Colchester, Oldham and Bury, while younger brother Tom, now at Preston, had a brief loan spell at Bradford in 2008.
Bradford boss Phil Parkinson had been looking to fill the void left by Andrew Davies, who signed for Scottish Premier League side Ross County in June.
