City come to the King Power Stadium on Saturday sitting eight points clear at the top of the Premier League.
Pep Guardiola's men have won 10 of their 11 outings this season and have scored 38 goals while Leicester sit 12th, four points above the bottom three.
Leicester, under Claudio Ranieri, won the corresponding fixture 4-2 in December last year and Puel believes they can succeed again.
He said: "It's a great opportunity to win, and for the first time, they could lose . I think we can have opportunities against this team.
"They have players of fantastic quality but the most important thing is to focus on our game and what we can do - not just to disturb them but to get a good result.
"Normally they are a team with a lot of possession - it's difficult to keep a lot of focus when you run after the ball and in the second half they can score.
"It's important for us to believe in our quality. It's important to think we have fantastic quality
"Pep Guardiola is a fantastic manager. It's a complete team and with Paris (St Germain) they are the best team in Europe at this moment.
"It's an exciting game for us, to play against a great team and a great manager.
"It's an opportunity for us to play good football and, I hope, get a good result. We want to see what we can do against this team. We respect them but we don't fear them."
Puel is again without Adrien Silva who this week saw his request to provisionally register for the Foxes rejected while the Court of Arbitration for Sport considers his personal appeal.
The midfielder currently cannot play until January after the paperwork for his £22million move from Sporting Lisbon in August arrived 14 seconds after the deadline.
Puel said: "It's difficult for him. It will be a good thing for him, for the squad, to have Adrien available. It's the regulations, it's a fact and we need to accept this and prepare Adrien for January."
Robert Huth (knee) and Matty James (Achilles) are also unavailable but Ben Chilwell (knee) and Vicente Iborra (ankle) are fit.
