Napoli, who last won the competition in 1989, will host FC Dnipro Dnipropetrovsk in the first leg before travelling to the Ukraine to play for a place in the final.
Fiorentina face the challenge of Sevilla, who could win the competition for the fourth time in a decade, with the first leg played in Spain.
Should the two Serie A sides make it through to the final it will guarantee the first Italian winner of the competition since Parma in 1999.
Napoli beat Trabzonspor, Dinamo Moscow and Wolfsburg to reach the last four, while Fiorentina conquered Tottenham Hotspur, Roma and Dynamo Kiev.
Dnipro have never made it this far in Europe before and made it through after knocking out Olympiakos, Ajax and Club Brugge.
Sevilla are the defending Europa League champions and have faced significant challenges so far, knocking out Borussia Monchengladbach, Villarreal and a much-fancied Zenit St Petersburg side.
The first leg of both ties will be played on May 7, with the return leg a week later on 14 May.
