Conference South side Bromley led after just a minute thanks to Louis Dennis and after Dartford had battled back to lead 3-2, Danny Waldren looked to have forced a replay when he equalised for the home side with 15 minutes remaining.
But Dartford, who are 21st in the Conference, advanced into the second round for the first time since 1984 thanks to Bradbrook's header nine minutes from full-time.
Bromley winger Dennis sent the Fortress Stadium wild when he put the home side ahead after a minute when he turned in Moses Ademola's pass.
The hosts kept that lead until half-time, but moments after the break Dartford's Danny Harris headed home Peter Sweeney's corner to level.
Ryan Hayes then gave Dartford the lead in the 56th minute, but almost immediately the home side were level when Ademola found the net following an error from visiting goalkeeper Jason Brown.
Dartford captain Elliot Bradbrook then put the visitors back in front with a header only for Waldren to once again bring the scores level.
But Dartford captain Bradbrook's brother, Tom, was on hand to head in an 81st-minute winner to send Dartford into the hat for the second round.
