The 28-year-old scored once in eight appearances for Porto last season as the club finished behind champions Benfica in the league and lost out in the Champions League Round of 16.
The Belgian is Huddersfield manager David Wagner's first signing in the current transfer window as the club return to the top-tier after 45 years.
"I am very happy that we have made Laurent our first signing of this summer window," Wagner told the club's website (www.htafc.com).
"He is a proper striker; one who has played for a very good Belgium side, featured in elite European competition and who has won domestic titles."
Depoitre previously won the Belgian league title and the Super Cup with Gent in the 2014-15 season. (Reporting by Aditi Prakash in Bengaluru; editing by Amlan Chakraborty)
