The London club said on their Twitter account that "further assessment & scans have confirmed that @Jan_Vertonghen sustained damage to his medial collateral ligament in his knee at Palace.
"Our medical staff will continue to monitor his progress during his rehabilitation to determine when he will be ready to return to training.
"Get well soon, Jan!"
Vertonghen, 28, suffered the injury after colliding with Connor Wickham in his side's 3-1 win at Crystal Palace in the Premier League on Saturday.
He left the pitch before undergoing a scan in Barcelona where Spurs are preparing for their FA Cup fourth-round tie with Colchester on Saturday.

There are doubts the player will be back for matches in the Premier League with Watford and Man City and a Europa League last-32 tie with Fiorentina. but the fear is the player could be missing for months rather than weeks with Spurs five points off leaders Leicester in the table as they chase a first English title in 55 years.
He is almost certainly out of the Colchester match and Norwich City in the Premier League, according to reports.
Vertonghen could face between three to 12 weeks out depending on the severity off the injury.
