Mertesacker, who was made club captain following the departure of Mikel Arteta last summer, had entered the last six months of his contract but will now remain at the club until the summer of 2018.
"He has an option that we have taken," Wenger said at his press conference on Thursday. "There was no negotiation, just an option that we have taken."
Mertesacker, 32, moved to the club in 2011 and has made 207 appearances for the north London club.
Wenger also revealed that the club would not be making a move for want-away West Ham forward Dimitri Payet.
"I rate Payet but I don’t need Payet because we have so many creative players," said Wenger. "We have many players who can play in this position."
