Kane limped off the pitch after receiving a knock to his right ankle in the tenth minute, and underwent scans on Tuesday to assess the damage, after waiting for the swelling to go down.
In a series of tweets on Tuesday night, the club confirmed the injury was similar to one he had sustained earlier in the season which saw him ruled out for seven weeks, though they added that the injury was less severe than the previous occasion.
The club added: "Harry will continue to be assessed by our medical staff while he undergoes rehabilitation at our training centre."
Since returning from his previous injury, Kane has enjoyed blistering form, enjoying player of the month award for February as well as a string of goals.
Currently the league's top scorer, jointly with Everton's Romelu Lukaku, Kane looked likely to better his tally of 25 goals last season, which saw him take the golden boot.
With just 10 league games remaining this season, and an FA Cup Semi-final against Chelsea in April, the injury comes at a crucial time for Spurs, pushing to improve on their standing at the end of last season, in third place.
Spurs are currently second in the league but level on points with third place Manchester City.
