The Minstermen secured the points, and only their second win in 11 fixtures, with three goals in 13 minutes either side of half-time.
Wes Fletcher struck just before the interval and further strikes from Michael Coulson and Jake Hyde soon made the game safe.
The result lifts York two points above the relegation zone, while Cambridge are seven points above the bottom two.
"We got everything spot on. It was near-on perfect," declared Wilcox after the game.
"Our game plan was good, making sure we were nice and solid. We could have scored more goals, it was a terrific performance.
Referring to his side's recent poor run of form, Wilcox added: "I do feel that the performance has been coming.
``We had that killer instinct this time and that was the difference. When the opportunities come you've got to put them in the net. We always speak about the first goal and how big it is. That was shown again tonight.
"I was really pleased with how we managed certain periods of the game, we looked after the ball and moved it around the pitch. That was what got us our second and third goals.
``We had a lot more legs in our team than they had. They're big and strong and powerful but leg-wise we had more poise.
"We aren't getting carried away. We know where we are in the league. We've moved up a couple of places but there's still a lot of work to do."
Cambridge boss Richard Money was at a loss to explain his side's performance after consecutive clean sheets in their previous two games.
"I certainly didn't see it coming," said Money. "I didn't expect it given the way we've played in the last two games, especially after Saturday (a 2-0 win at Morecambe). But it was a poor performance and we ended up getting what we deserved."
Speaking on the first goal just before half-time, Money reflected: "It's a poor goal. It's another mistake and we have to say that we've been like that all season.
``The facts are that we haven't defended well enough all season in this league and that's why we are where we are in the table.
"If you take half-time out of it then it's three goals in just over 10 minutes and it would appear at the minute that we either keep a clean sheet or we concede three goals in 10 minutes.
"If you can't play with any stability then the lack of confidence spreads through the team and you end up with a performance as disappointing as it was."
