The 2010 Olympic and four-time World champion came home in 1:28.638 minutes to push Canadians Kimberley McRae and Alex Gough into second and third respectively.
Hufner's record-equalling victory means she also takes the lead in the overall World Cup standings on 240 points. Her teammates Natalie Geisenberger, who finished fifth in Lake Placid, and Dajana Eitberger are in second and third with 225 and 209 points respectively.
But despite missing out on top spot here there was more for McRae to celebrate as she was part of the Canadian quartet that won the team relay in Lake Placid.
McRae, Sam Edney, and the doubles pairing of Tristan Walker and Justin Snith took victory, with Russia second and hosts America in third.
Defending champions Germany – with the team of Hufner, Felix Loch and the doubles pairing of Toni Eggert and Sascha Benecken – had to settle for seventh place.
