The 34-year-old, an eight-time world champion and 2010 Olympic gold medallist, sped to success in Canada after clocking the first and third fastest times of the competition on the day with her two runs.
She amassed a total time of 1:33.442, 0.115 clear of her nearest rival – home favourite Alex Gough, a six-time world championship medallist herself, who had to settle for silver in front of her home support.
Huefner's compatriot Natalie Geisenberger, who has two Olympic golds from 2014 and seven world titles, finished third with a combined time of 1:33.603.
Geisenberger leads the overall standings with 440 points after five rounds, with three victories and two third-place finishes under her belt in what has been a terrific campaign to date.
Huefner is her nearest challenger on 400, with Gough third on 283, ahead of the World Cup's sixth round in Lake Placid.
