Dan Evans, the last remaining Briton in qualifying, went down 7-6 (6) 6-2 7-5 to Japan's Yuichi Sugita in the final round of the Roehampton event.
Winning a tight first set proved crucial for Sugita, who then quickly took control of the second.
Evans, a key member of GB's Davis Cup team in 2012 and 2013, fought hard in the third but losing his serve at the end of the contest saw him beaten in two hours and 14 minutes.
The host nation will still have plenty of players in the men's singles at the All England Club next week.
Andy Murray heads the British list with Aljaz Bedene also in as of right. Kyle Edmund, James Ward, Liam Broady and Brydan Klein have all been handed wild cards.
In the ladies' singles, Heather Watson was the only Briton to make the automatic field cut-off. She will be joined by wild cards Laura Robson, Johanna Konta and Naomi Broady.
