Both men struggled with injury during the French Open. Tsonga was 5-2 up on Ernests Gulbis in his third-round match when he withdrew in tears because of a thigh problem.
That cost the Frenchman his place in the top 10 and he has been unable to recover in time to play at Queen's, where he reached the final in 2011.
Tsonga said: "I am very disappointed that this injury means that I will not be able to play in the Aegon Championships at the Queen's Club.
"It is one of my favourite tournaments, home of some very happy moments for me on the court - reaching the top 100 for the first time and playing in the final - and I was looking forward to coming back. I hope to return in the future."
Nadal pulled out last week due to the wrist injury he suffered at Roland Garros.
Defending champion Andy Murray will be the top seed for the tournament, which begins on Monday and is broadcast live on Eurosport.
