Since losing in the semi-finals of Wimbledon to Venus Williams, Konta, seeded fifth in the tournament in China, has been on an awful streak of form, losing five of seven matches.
Those two wins both came in Cincinnati, and Konta has now lost in the first round of the US Open and WTA tournaments in Toronto, Tokyo and Wuhan since the start of August.
Barty demolished Konta in the first set, and while Konta fought back to take the second and served for the match but her run of poor forms continues with world number 37 Barty dominating the tie-break to progress.
There were also defeats for US Open finalists Madison Keys and champion Sloane Stephens – as well as former world number one Angelique Kerber, as some big names fell.
Stephens, seeded 14th, went out 6-2 6-2 to Wang Qiang of China. "I didn't play a great match, but my opponent played very well," said Stephens. "Sometimes you have those days. So it's just not a good day today."
Keys was defeated 6-2 7-6(4) by Varvara Lepchenko while Kerber fell to France’s Caroline Garcia despite winning the first set, 3-6 6-3 6-1.
Elsewhere in Wuhan, ninth seed Agnieszka Radwanska beat another Wimbledon semi-finalist, Magdalena Rybarikova, 6-4 7-5.
The Pole was a break down in both sets but came through in over two hours to take her place in the second round.
