It was Murray's 11th appearance at Queen's Club and the 31-year-old finally got his hands on the famous trophy after he and Brazilian partner Soares bested Julien Benneteau and Edouard Roger-Vasselin 6-2, 6-3.
He became the first Brit to win doubles at Queen's since Jeremy Bates in 1990, although the victory came in slightly bizarre fashion.
Before they could focus on the final, Murray and Soares had to complete their rain-delayed semi-final against Marcin Matkowski and Marin Cilic, just minutes after the latter had lost the men's singles final to Feliciano Lopez.
The British-Brazilian pair secured the point they needed to level the match at one set each before winning the Champions tie-break – triumphing 1-6, 6-3, 10-8.
And they maintained that form in the final to cruise past their French opponents, leaving Murray ecstatic to clinch a maiden Queen's title.
"Eleven years is a long time but I wasn't always playing at the top of the game, so I was scrapping to get into this tournament," explained the Scot.
"It's nice to not have to worry about that anymore. Thanks a lot to Bruno for an amazing week and we played some amazing tennis.
"Today was really weird and I think my heart-rate dropped below 180 for the first time when we won the first set in the final having had to get through the semi-final before.
"We played a great match, it was a long day but it was worth it."
It was Murray and Soares' second doubles title in as many weeks after triumphing at the Mercedes Cup in Stuttgart seven days ago – beating Oliver Marach and Mate Pavic in the final.
They are in fine form heading into Wimbledon – which gets underway a week on Monday – and Murray is feeling positive.
"We've had two great events, in Stuttgart last week and now here," he added. "These are the best grass courts and we couldn't ask for more.
"The crowd were awesome. It's great to come out here and play the final in front of so many people – it's a lot more fun than playing on the back courts and great to have that support."
