In the evening session, Roger Federer breezed through 6-1 6-2 6-1 win against Steve Darcis on Arthur Ashe, but there was a huge shock in the women's draw as last year's runner-up Caroline Wozniacki crashed out to Petra Cetkovska.
Murray rallied from two sets down in the heat and humidity of New York to see off a spirited challenge from the Frenchman.
For two sets, the French challenger confounded Murray with brilliant shotmaking and smart serving.
The uncompromising aggression was simply too much for Murray to handle, as the diminutive Frenchman tore holes through Murray's famous defence. Such was Mannarino's dominance, by the time the third set rolled around, he had accumulated ten times as many forehand winners as Murray.
However, the Scot kept his composure and waited the Frenchman out. As the third set came around, the searing heat and humidity began to take its toll on the smaller man. Whilst the challenger hit the wall, the former US Open champion soared and never looked back.
In each of the final three sets, the Scot broke his opponent's serve within Mannarino's first two service games of each set. It was as convincing a recovery from such a score as can be, and he happily marched to victory.
Murray will face Brazilian clay court specialist Thomaz Bellucci in the third round.

Federer took just 80 minutes to dispatch the 66th-ranked Darcis under the lights at Arthur Ashe Stadium and has lost just nine games in his first two matches.
In an ominous warning to his rivals, Federer said his early form points to a successful fortnight at Flushing Meadows, one of his favourite haunts.
"Very often I start this tournament quite strong," said the 34-year-old. "It's always gone quite well for me.
"I've always enjoyed conditions here - the balls, the speed of the court, the atmosphere in the arena. I think this is another good year."

Federer won five US Open titles in a row from 2004 to 2008 but has not made it to the final in New York since finishing runner-up in 2009.
The evergreen Swiss is feeling confident this time after a strong season and impressive build-up to the event.
He was a finalist at Wimbledon in July and has captured five ATP titles this year, including last month's Cincinnati Masters, where he defeated Novak Djokovic in the final.
Playing with free-flowing aggression, Federer fired 11 aces and a total of 46 winners past Darcis while committing 25 errors.
Comfortable in the New York spotlight, he matched Serena Williams for the most US Open wins at night, improving his record to 29-1.
Federer's next opponent is Germany's Philipp Kohlschreiber, a straight-sets winner over Czech Lukas Rosol. Kohlschreiber was one of 13 seeds to make it through to the next round on Thursday, with Ivo Karlovic and Jack Sock the day's main casualties.
The Croatian was beaten by Jiri Vesely, while home favourite Sock was forced to retire despite being a set up on Ruben Bemelmans as he collapsed on court due to the heat and humidity.
Elsewhere Donald Young was victorious over Brit Aljaz Bedene, while Leyton Hewitt waved goodbye to Flushing Meadows after he was beaten 7-5 in the fifth set by compatriot Bernard Tomic.
"It was nice to be able to turn it into a decent match," Hewitt, who initially fell two sets behind, said afterwards. "Tennis has given me this great life and that's the best thing.
"I will pass on stuff to the young guys. That's my next role, to help those boys out."

In the women's draw, Wozniacki became the highest seed to fall in the upset-filled women's draw when Cetkovska stunned the Dane 6-4 5-7 7-6(1).
The upset left only three of the top 10 seeds still alive in the year's final grand slam - number one Serena Williams, who is seeking the fourth women's calendar-year Grand Slam, second seed Simona Halep and fifth seed Petra Kvitova.
Cetkovska, who has tumbled to a 149th ranking after missing seven months with a hip injury before returning in March, was playing in the Open thanks to a special injury exemption.
She rode a powerful forehand that produced 29 of her 61 winners to victory over Wozniacki, saving four match points held by the Dane - all on winners off her racquet.
"I said it's now or never, so I just went for it," she said in an on-court interview about her string of match point saves. "It's just amazing."
"So happy to be able to be back and to compete," added the 30-year-old Cetkovska, who will play Italy's Flavia Pennetta, a straight sets winner over Romanian Monica Niculescu.
Wozniacki was not the only seed to fall on Thursday, with Wimbledon finalist Garbine Muguruza beaten by Britain's Johanna Konta. Elsewhere there were victories for Simona Halep, Petra Kvitova, Angelique Kerber and Victoria Azarenka.
