The Russian was placed second after the first run on the Vancouver 2010 Olympic track, but produced a near faultless second run to jump up the leaderboard and take the gold medal.
Behind him Lamin Deen drove to a second consecutive four-man medal for Great Britain, improving on Brad Hall's bronze from Park City.
Deen stopped the clock in 50.66 seconds in the first run, a track record at the fastest sliding centre in the world, but could only manage the 11th fastest run at the second attempt, meaning he had to settle for silver.
"I've been in this sport nearly 25 years and this weekend is right up there with anything I've experienced," said GB Bobsleigh head coach, Lee Johnston – whose other sled, drove by Hall, finished ninth.
"It's on a par with last week - and to have two weeks like that in succession is pretty special.
"In two days of racing in Whistler we've won a silver medal; broken a long-standing track record, had two sleds in the top 10 again and seen Brad equal his best 2-man finish of his career.
"The level keeps lifting."
Nico Walther took bronze behind him, nearly half a second down on Kasjanov and two tenths of a second behind Deen.
Justin Kripps guided Canada into fourth with Johannes Lochner in fifth, meaning the German still leads the overall World Cup standings, with Walther second, Kripps third and Deen in fourth.
