After finishing third in Sunday's 20km time trial, took a 14-second overall lead over Liberty Seguros' Jorg Jaksche and the rest of the field.
Three flat stages makes it much harder for the German and the likes of two-time Giro winner Paolo Savoldelli to catch Dekker.
Savoldelli currently lies fourth, 40" back of the Dutchman, who shrugs off the pressure that comes with the leader's jersey.
"I came here to win, I've prepared for that and I know I can count on [Michael] Boogerd and [Juan Antonio] Flecha," Dekker told L'Equipe.
"So whether or not we climb the San Giacomo, it's all the same to me."
