The striker limped off with less than 10 minutes gone at White Hart Lane after appearing to turn his right ankle.
In bad news for England manager Gareth Southgate, it was the same ankle he turned in a match against Sunderland in September - a problem that ruled him out for six weeks.
Kane would later leave the ground on crutches and wearing a protective boot.
Southgate names his squad next week ahead of the friendly against Germany on March 22 and the World Cup qualifier with Lithuania four days later.
