A first-half Marcelo double followed by goals after the break from Memphis Depay, Bertrand Traore and Mariano Diaz powered Lyon on to 63 points from 32 games. They now lead Marseille, who were unable to break the deadlock against Montpellier at the Stade Velodrome, on goal difference with six games left.
Second-placed Monaco, who beat Nantes 2-1 at home on Saturday, are on 70 points with runaway leaders Paris St Germain on 84 after they were held 1-1 at St Etienne on Friday.
PSG will be crowned champions if they beat Monaco at the Parc des Princes next Sunday.
"It was the perfect start for us," said Lyon coach Bruno Genesio after Marcelo opened the scoring in the first minute, the Brazilian doubling the tally on 20 minutes.
Depay put on a sterling display and made it 3-0 in the 65th minute with a delicate chip, before setting up Traore and Diaz for the final two goals.
