The Belgium international passed his medical with the North London club after the two clubs finally agreed on a fee following prolonged negotiations.
Vertonghen is the club's second signing of the summer following Gylfi Sigurdsson and arrives at a club lead by a new manager in Andre Villas-Boas.
The 25-year-old helped Ajax win a second consecutive league title last season and scored 10 goals in 37 games before being named Player of the Year.
Tottenham confirmed the news on their website, writing: "We are delighted to announce that Jan Vertonghen has completed his transfer from Ajax after successfully passing his medical."
Vertonghen emerged from the famed Ajax academy and made his first-team debut in the 2006-07 Eredivisie season.
He spent a spell on loan with RKC Waalwijk during the same season and leaves Ajax having made 220 appearances for the Amsterdam club.
