James Chester, Matt Smith, Tom Lawrence, David Brooks and Tyler Roberts come into the side in place of Chris Gunter, Declan John, Ethan Ampadu, Aaron Ramsey and Sam Vokes.
The away team are coming into the match on the back of two defeats - to Denmark and Spain - but will hope for a repeat of the 4-1 win in Cardiff over Martin O'Neill's men in the same competition.
Giggs is without the services of star men Gareth Bale, who is fatigued, and Aaron Ramsey, who became the father of twins on Sunday.
The Republic of Ireland team is as expected with Callum O’Dowda missing after he had spells of dizziness following a collision last week.
Ireland: Randolph; Doherty, Duffy, Keogh, Christie; Robinson, Arter, Hendrick, McClean; O'Brien, Long
Wales: Hennessey; C Roberts, Williams (C), Chester, Davies; Allen, Smith; Brooks, Wilson, Lawrence; T Roberts.
