Irishman Harrington, who birdied the par-five 18th hole, again had to settle for the bridesmaid's role in the elite four-man event, having lost to Argentina's Angel Cabrera in a play-off last year.
The pair had finished the 36 regulation holes on four-under-par 136 at The Mid Ocean Club in Tucker's Town, Furyk matching Harrington's two-under 68 with a birdie at the last.
Harrington, who set the pace with Furyk in Tuesday's opening round, held a two-stroke lead with six holes remaining before double-bogeying the par-four 15th.
Furyk had cut the deficit to one with a birdie at the par-three 13th and edged a stroke in front following Harrington's stumble on 15.
Although the American again slipped one behind when his bogey at the par-three 17th was trumped by Harrington's birdie, he forced the play-off with his birdie at the last.
"I'm very happy with the outcome," a beaming Furyk said after winning the title for a second time.
"I really came here with, I won't say low expectations but not a lot of expectations. I have not played a lot of golf since the (September 25-28) Tour Championship."
"It's a wonder I'm even in the event," he added. "So I kind of treated the week like I had nothing to lose, was going to come over here and have fun.
"I kept plugging away and didn't let too many things bother me. I didn't really worry about how anyone else was playing and that all worked out in the end."
Twice US Open champion Retief Goosen carded a 71 to finish third at one-over 141. Fellow South African Trevor Immelman, the US Masters winner, was a further four strokes back after a 69.
The event, held in Bermuda for the second consecutive year, traditionally features the winners of the season's four majors.
Seven-times champion and world number one Tiger Woods is a conspicuous absentee, having ended his campaign after his US Open victory in June to have reconstructive knee surgery.
With Harrington having won The Open and the US PGA Championship, Goosen and Furyk were late additions.
