De Klerk started in the No. 9 jersey against France in Paris last weekend as the Springboks won 29-26, but he missed South Africa's first November tour match against England because the encounter fell outside World Rugby's Test window.
He will play for struggling Sale away against champions Saracens in the English Premiership.
The Springboks have three other scrumhalfs available for the test against the Scots.
Ivan van Zyl started against England, while Embrose Papier was on the bench against England and France.
Uncapped Louis Schreuder is the other scrumhalf in the Bok squad with coach Rassie Erasmus to name his team on Thursday. (Writing by Mark Gleeson; Editing by Hugh Lawson)
