Ennis celebrated her 25th birthday on Friday and she claimed herself the perfect belated present, romping home in 7.97 seconds to hand the Great Britain team five points.
It wasn't quite the British record of 7.95 that Ennis clocked last year but it was still good enough to beat Jones, who paid for the price for hitting one of her hurdles.
"I love competing here," said Ennis. "It's a quick track and it's great to be able to run against some really good girls so I'm really happy with that.
"I had to do my captain's duty earlier but I had enough time to get my head ready for the competition and it went really well."
Olympic bronze medallist David Oliver of the USA took victory in the men's event, clocking 7.51 for his second consecutive win in Kelvin Hall.
Oliver was sluggish out of the blocks but he swiftly got into his stride and ensured Great Britain's Andy Turner, who finished in 7.66, had to make do with second place.
"I was just trying to work on my technique," said Oliver. "It felt pretty good because I only got here 24 hours ago but I came here to represent and I feel that I did that.
"I'm just trying to stay consistent this season. When you put your feet in the starting blocks it means you are ready and so you have to represent and I'm glad with home I did that."
Commonwealth and European champion Turner admitted he will need to raise his game if he is to live with the likes of Oliver across the season.
"It was a good race to the first hurdle but over the second and third I didn't push hard enough and if David gets a lead he's not going to let it go.
"Time-wise I'm pretty happy but performance-wise I am not."
