Australian Hosking prevailed as the sprinters took charge on the 114-kilometres stage from Montagnana, winning ahead of Marta Tagliaferro (Ale-Cipollini) and Giorgia Bronzini, her Wiggle-High5 team-mate.
Evelyn Stevens (Boels Dolmans) retained the overall lead as the top of the general classification was unchanged.
"It was unexpected for me, I'm shocked," Hosking said.
"I really don't like the narrow curves close to the finish but the team did great work and I must thank my team-mates."
Stevens leads by two seconds overall from Katarzyna Niewiadoma (Rabo-Liv) and her Boels Dolmans team-mate Megan Guarnier is 18 seconds behind in third.
Tuesday's fourth stage has been altered due to a landslide on the west coast of Lake Iseo.
The route from Costa Volpino to Lovere now includes a climb in Pianico 3km from the start, but is expected to end in a sprint before the big mountains.
