The 24-year-old, who won her first two Olympic golds at the venue during London 2012, married six-time Olympic champion Jason Kenny in September.
"I'm really excited to be back racing again," Kenny said.
"I needed a break after [the Olympics] but I'm looking forward to getting back on my bike and enjoying racing after the pressure of Rio.
"I know the others won’t make it an easy race back for me, but it’ll be great fun and even better it will be in front of a home crowd.”
Fellow Olympic gold medallist and Matrix Fitness RT team-mate Elinor Barker will join Kenny in London and will also race at Round 1 in Manchester.
Dame Sarah Storey announced last week she will return to racing for the first time since the Rio 2016 Paralympics, riding as team leader for championship leaders Podium Ambition p/b Club La Santa at Round 1 in Manchester.
