John Still, who this week celebrates two years as manager, remains disappointed by the Hatters' defending in the defeat at AFC Wimbledon last weekend so Wilkinson, available again after serving a two-match suspension, may resume at centre-half.
Tyler missed the Dons defeat with a knee injury but he looks set to replace Elliot Justham in goal on Saturday.
Winger Ryan Hall trained on Thursday following a bout of tonsillitis and could come back into contention but captain Stephen McNulty will complete a four-game ban while Alex Lawless (foot), Pelly Ruddock Mpanzu (hamstring), Paul Benson (broken leg) and Nathan Doyle (knee) remain out injured.
Accrington boss John Coleman has to decide what to do over his formation after utilising a 3-5-2 to good effect in midweek.
They recorded a 1-0 win over Burton in the process so Kal Naismith and Anthony Barry will hope to hang on to their places.
Coleman has a "healthy squad" but will check on the injured Nicky Hunt and fellow full-back Adam Buxton, who is struggling with a bout of illness, and their availability may sway his thinking on how they set up.
Midfielder Matt Crooks, a former Stanley loanee, will hope to make his first start as a permanent Accrington player.
