The England striker missed the clash with Manchester United due to a hamstring injury but he returned for the victories against Real Madrid and Crystal Palace.
However, against Palace he took a knock on his ankle that saw him withdraw from Gareth Southgate's squad for the friendlies against Germany and Brazil.
There were concerns he could miss the derby but Pochettino confirmed at his book launch that Kane would be fit to face Arsenal.
"Harry Kane is ok. You can see in the last game he received a kick to his knee but he will be available for the next game," said the Argentine.
Following the win over Palace Spurs are third in the league, eight points behind leaders Manchester City but four points ahead of sixth-placed rivals Arsenal.
Spurs won the last derby at White Hart Lane in April but the previous three matches were all draws.
