The Briton won 6-4 6-2 to upset to world number three Radwanka and add to the Stanford Classic title she won in July 2016.
The victory serves as perfect preparation for Konta ahead of the Australian Open, and the 25-year-old will start the tournament ranked nine in the world.
"I was born here, so this is a very special moment for me," Konta said after receiving the trophy. "I'm actually really happy."
Belgium's Kirsten Flipkens will be Konta's first-round opponent in Melbourne, while she could potentially face Serena Williams in the quarter-finals.
Konta simply overpowered Radwanska in Sydney, hitting 18 winners on her way to taking the first set, claiming the all-important break in the third game.
The British number one then broke Radwanska twice to race to a 4-0 lead in the second set, before closing out the match after one hour and 23 minutes on court.
"Well done Jo, it was unbelievable tennis today, I couldn't have played any better," said her opponent.
