Coach Artur Jorge called up the uncapped French-based defender to a 22-man squad for a match in Yaounde on October 9 which Cameroon must win to make sure of an African record sixth World Cup finals appearance.
The 21-year-old Assou-Ekotto was born in France but has a Cameroonian father.
Chelsea wingback Geremi will miss the match through suspension after picking up a yellow card in Cameroon's last World Cup qualifier, a dramatic 3-1 away win over the Ivory Coast in Abidjan.
The result resurrected Cameroon's chances of qualifying, lifting them to the top of Group Three of the African zone qualifiers. They lead the Ivorians by one point.
If Cameroon fail to win, the Ivorians can still qualify by beating Sudan away on October 8.
