Young has been troubled by an ankle injury and missed both Rio Ferdinand's testimonial on Friday and today's Community Shield victory over Wigan at Wembley.
With little chance of being fit to face the Scots, Young has now returned to his club for further treatment ahead of the opening weekend of the Premier League season.
"Manchester United forward Ashley Young has withdrawn from the England squad," said the FA in a statement.
"Following discussions between England medical staff and their counterparts at the club, it was clear Young would not be ready for the match with Scotland on Wednesday."
No replacement has been called up, although as both England seniors and Under-21s are at St George's Park and are being managed by Roy Hodgson for their respective games against Scotland this week, finding one would not be a problem if the remaining 24 players needed it.
