Drinkwater, 27, becomes Chelsea’s sixth signing of the window after moves for Willy Caballero, Antonio Rudiger, Tiemoue Bakayoko, Alvaro Morata and earlier on transfer deadline day Davide Zappacosta.
The England international signed a five-year contract at the Stamford Bridge club.
“I’m delighted to be a Chelsea player and can’t wait to get started,” the midfielder told the club’s official website.
“It has been a long journey to get here but I am very happy and am looking forward to helping the club win more trophies.”
Drinkwater, who came through Manchester United's youth-system, won three England caps since making his international debut in March 2016.
Another piece of late deadline day business saw the Premier League champions also fork out £23m for Torino defender Davide Zappacosta.
