Qualifier Swail, the world number 45, followed up his 5-3 victory over Mark Allen on Thursday with a solid display against Murphy.
He lost a scrappy opener, but runs of 70, 44, 37, 52 and 47 helped him establish a 4-1 lead.
Murphy won the sixth frame despite an opening Swail knock of 52, but the roles were reversed in the seventh frame as Swail recovered from his opponent's break of 53 to seal the match with a superb double on the pink to a middle bag.
Swail will face Marco Fu in the last eight on Saturday.
"I'm over the moon with that, it was another tough match, but I didn't want today to be an anti-climax after the Mark Allen victory yesterday," said Swail.
"I just wanted chances and luckily I got offered them after the first frame and I took them.
"I missed a few silly balls at 4-1 up and that let him back in but I was always in control of the match. Funnily enough, I felt edgy at the start but the 70 break in the second frame settled me down a lot."
Mark Selby enjoyed a 5-3 victory over seven-times world champion Stephen Hendry.
Selby moved 2-0 clear with breaks of 76 and 105 before Hendry made it 2-1 with a 71. Selby moved 3-1 clear with a 83 only for Hendry to level at 3-3 courtesy of runs of 72 and 60.
Hendry missed key balls in the next two frames as a break of 61 helped the former Masters champion Selby over the winning line.
"We both played well and didn't do a lot wrong, it came down to nerve in the seventh," confessed Selby. "I'm delighted to get through as I've been struggling a bit recently," said Selby. "Games against Stephen are always special as he's a legend of the game, but I concentrated on my own stuff and it worked out well."
Ding Junhui continued his quest for back-to-back Masters titles after his success at Wembley last month with a 5-2 win over Peter Ebdon.
Anthony Hamilton could not build on his surprise 5-4 win over world champion Neil Robertson in the first round.
The Nottingham break-builder went down 5-2 to 2006 world champion Graeme Dott, who was delighted to keep his concentration after the sad news that his friend and practice partner John Higgins's dad had died earlier in the day.
