The Rocket faces the Belgian champion in the first round of the event showcasing the new format of the game, which takes place at the Indigo2 Arena on October 30.
The other clashes at the eight-man knockout tournament see Ding Junhui take on wildcard Jimmy White, Masters champion Mark Selby meet Ali Carter and current world champion Neil Robertson play former number one Shaun Murphy.
"I was hoping to get one of the more well-known lads as I don't think Power Snooker will suit them," O'Sullivan said.
"Luca is a real wildcard and I think he will be waiting to put me on my backside."
Brecel said: "I was delighted to be one of the eight players to get into Power Snooker, and to go head to head with Ronnie is fantastic.
"This is not traditional snooker, this is a real shoot out and I think I can beat Ronnie. Power Snooker is a young man's game."
The first knockout round will be played in the afternoon session in east London with the evening session featuring the two semi-finals and the third place play-off before concluding with the Grand Final.
