The 2005 world champion - who lost this year's final to Stuart Bingham - made knocks of 102, 69, 65, 52, 58, 92 and 70 to secure a meeting with Zhou Yuelong in the last 64 of snooker's second biggest ranking event.
Murphy said he was impressed with Hugill, 21, who conceded he failed to make the most of the chances that came his way in the opening frames.
“I played well,” said Murphy. “I didn’t really give Ashley much chance to get going because, with the new kids, you never know what you are going to get. I remember seeing him when he was a young boy. He spent a bit of time with my old coach and he can definitely play.
“It is good to see another young player from this neck of the woods trying his hand. I hope he goes from strength to strength.”
In the evening session, Judd Trump moved into the last 64 as he beat Hammad Miah 6-1 - without making a century break in the match.
He will now face Stuart Carrington.
“A lot of people have raised their games against me this season but it has still not been bad,” Trump said. “I have had a couple of early defeats but I feel like I am playing well again now and I have practised hard for this one.
“I have had a cold for the last few days but I have a three day break now so I should be fine for the next game.”
There was a shock elsewhere, however, as 2003 winner Matthew Stevens was dumped out 6-5 by Chris Wakelin - who overcame some nerves to seal the deciding set on the final black.
“When it went 5-5 I turned to jelly," Wakelin admitted. "But I’m learning to deal with these situations and gaining experience with matches like that. I’m really looking forward to the next one now.”
