The two Australians are again expected to be the leading protagonists for the championship, although an impressive performance from former MotoGP star Max Biaggi suggested his Suzuki will be competitive in 2007.
Corser clocked 1:59.1 at the Losail circuit on his Yamaha R1, two tenths of a second ahead of Superbike rookie Biaggi.
Having set the pace on the first day of two, Noriyuki Haga was unable to better his time of two minutes exactly, while Biaggi's team-mate Yukio Kagayama's best lap was 2:00.3.
Honda's Australian duo Karl Muggeridge and Josh Brookes were both three seconds off the pace.
Corser is bidding to reclaim the title from Ducati star Bayliss, who ripped around the Vallelunga track in an unofficial lap record 1:38.5.
The circuit is a new addition to the world championship, and the 37-year-old Bayliss said the test provided valuable experience.
"I guess the most important thing for us was to actually come here because it's the only place on the calendar this year that I haven't seen," he said.
His colleague Lorenzo Lanzi impressed on home tarmac, coming in just a tenth slower despite battling a bout of bronchitis.
The Airwaves Ducati British Superbike team were also present, with Leon Haslam and Gregorio Lavilla registering 1:39.3 and 1:39.6 respectively.
The 2007 season starts in Qatar, with the opening races taking place on 25 February.
