Murray fended off the Frenchman to come through 6-3 7-6(6).
The 29-year-old could usurp the Serb as early as November 7, although that scenario would rely on him winning the Paris Masters title next week and his rival crashing out before the final.
Murray looked on course for a routine victory after wrapping up the opening set and breaking immediately in the second, but Tsonga suddenly found his groove and broke back to force a tie-break.
The pair were evenly matched in the breaker, but some sublime hands from Murray at the net created a championship point – one he took with aplomb by firing down an ace.
The Brit’s charge was aided by a semi-final walkover against David Ferrer on Saturday, while Tsonga was pushed to a third set tie-break by Ivo Karlovic.
Murray has been hugely consistent in 2016, winning Wimbledon and the Olympic Games – albeit the latter yielded no ranking points – and reaching finals at the Australian and French Opens.
