Bubka, who is ranked only 321 in the world but who was given a wild card into the event, justified his selection with a surprisingly easy 6-0 6-3 win over Daniel Gimeno-Traver.
Tsonga was similarly dominant as he destroyed Ruben Ramirez Hidalgo 6-2 6-0.
Defending champion Nikolay Davydenko also had a textbook start with a 6-1 6-4 victory over Fabio Fognini of Italy.
"For sure it is nice to be back in Doha," said Davydenko.
"The New Year is important for me to win matches and get my confidence back to return to the Top 10. I won here last year and I like this tournament. If I have good results, why not come back here again and again. Now I have no injury, it is important to play well again."
World number one Rafael Nadal enters the tournament on Tuesday with a clash against Karol Beck, while Roger Federer is also competing in the tournament.
