The Spaniard secured her place in the last eight for the fourth time in Paris in the most comfortable fashion imaginable after Tsurenko sustained a leg injury.
The 24-year-old, who won at Roland Garros in 2016, did not have to exert herself at all with the Ukrainian unable to continue beyond her second service game.
Muguruza will next take on Maria Sharapova, seeded 28th in Paris, in the last eight.
The pair are on the same side of the draw as top seed Simona Halep, who remains the favourite to win her first Grand Slam after Serena Williams also retired through injury.
Tsurenko, ranked 39th, stretched to reach a Muguruza serve in the second game and immediately clutched the back of her thigh before calling for treatment.
A physio was summoned and Tsurenko was given extensive treatment.
She played on for five more points but walked to the net and shook hands after Muguruza held serve.
Muguruza is yet to drop a set in the tournament and will face twice champion Maria Sharapova for a place in the semis after the Russian's opponent, Serena Williams, pulled out before the start of their eagerly-awaited duel due to injury
