The Serb secured his progress through to the last 16 with a routine 6-4 6-4 6-4 triumph over Kokkinakis, who could not match his opponent's intensity at crucial times in the match.
The 19-year-old Australian fought valiantly to stay in each set, but Djokovic always had too much for him and booked his place in the second week at Roland Garros.
Djokovic, bidding for his maiden French Open title, hurtled towards a potential quarter-final clash against defending champion Rafa Nadal with what was an immaculate display against Kokkinakis.
Djokovic will await the winner between France's home favourite Richard Gasquet and 15th seed Kevin Anderson from South Africa.
