The brothers clinched a personal-best seventh title of the season and their 21st career title as a team in a title match lasting three hours, ten minutes.
Rain interrupted the match when the Bryans held set point against serve to level the match at one set all.
The Bryans converted the set point when play resumed and went on to become the first team since Stefan Edberg and Anders Jarryd in 1985-86 to win consecutive titles.
"It's unbelievable. This is our first major title of the year - we didn't win a Grand Slam. It feels great to snag a big one," Mike Bryan said.
Bob Bryan added: "It's a good year now. Seven titles is awesome - our best before this had been five.
"To come here on what is like our home court and the fans behind us is huge for us. For doubles it's the biggest stage there is.
"The spotlight is on the doubles and they pack the place out. We're stoked."
