Edmund, 21, needed four match points to finally defeat Czech opponent Vesely - eventually clinching a 6-4 5-7 7-6(6) victory that required two-and-a-half hours to complete.
That means he will now face Djokovic on Friday, as the Serbian gets his tournament underway.
"Very happy to come through that," Edmund told Sky Sports.
"Last week I made a lot of errors, especially in the areas I needed to be tough ... as the match went on today I was pleased that in the tough moments I was a lot tougher."
On the prospect of facing Djokovic, Edmund said he was looking forward to the task.
"It will be a huge honour for me to play the world No. 1," Edmund added. "He is the guy to beat at the moment, he is playing at such a high level.
"I watch him, watch his games a lot and learn from him ... it will be tough but more importantly it will be a great opportunity to see where my game is at."
Elsewhere on Wednesday there were victories for promising German teenager Alexander Zverev (over Michael Mmoh) and Inigo Cervantes, who had too much power for wild card entrant Andrey Rublev.
