Petrova didn't take a good start for her grass-court tournament, struggling against last year's runner-up in Birmingham before finally losing in three tensed set.
Shinobu Asagoe will next face French teenager Tatiana Golovin, after the 16-year-old went through thanks to Daniela Hantuchova's respiratory illness.
Serb Jelena Dokic, the 2002 champion, lost to American qualifier ShenayPerry 6-4 7-6 (7-4).
The American's next opponent is another Japanese player, Saori Obata, who defeated Kelly McCain 7-5 7-5.
Defending champion Magdalena Maleeva from Bulgary won her sixth straight game in Birmingham against Germany's Anca Barna 3-6 6-4 6-3 to set up a quarter-final clash with Thai Tamarine Tanasugarn, who defeated U.S.'s Jill Craybas 6-1 7-5.
Two other seeds were also dumped out of the tournament: France's Nathalie Dechy who lost to Italian qualifier Roberta Vinci 6-0 4-6 7-6; and Greek Eleni Daniilidou, who retired of her match against Japan's Akiko Morigami.
In the meantime, Russian Maria Sharapova and Frenchwoman Emilie Loit, twice a winner in 2004, both went through.
