The Colombian striker, who scored twice in the first leg, will not feature at the Stade Louis II Stadium after failing to recover in time from a hip injury he picked up against Bordeaux on Saturday.
City have a 5-3 lead going into the game, but have still named and attacking line-up with four changes from the team that beat Middlesbrough 2-0 in the FA Cup quarter-final on Saturday.
Willy Cabellero will start ahead of Claudio Bravo in goal, with Aleksandar Kolarov, Fernandinho and Bacary Sagna also recalled.
City XI: Caballero, Sagna, Stones, Kolarov, Clichy, Fernandinho, Sterling, Silva (C), De Bruyne, Sané, Agüero. Subs: Bravo, Zabaleta, Navas, Delph, Otamendi, Yaya Touré, Iheanacho.
Monaco XI: Subašic; Sidibé, Jemerson, Raggi, Mendy; Fabinho, Bakayoko, Silva, Lemar; Germain, Mbappé.
