Retaining the experienced duo will help new manager Bryan Robson stabilise the squad following the Blades relegation last season.
Winger Gillespie, 32, was a key part in the club's automatic promotion up to the Premiership in the 2005/06 season, and was one of their best performers during the last campaign.
Despite 35-year-old Lucketti only making eight league starts for the Blades since arriving at the club last season, boss Robson was keen to retain him.
Robson said: "Chris Lucketti and Keith Gillespie have signed new deals.
"It's done and dusted - Chris has signed a one-year contract and Keith has signed a two-year deal.
"I'm delighted about that as it keeps the squad nice and strong."
Robson was also keen to stress that striker Danny Webber will not be leaving the club.
The 25-year-old has been linked with a move to Hull City, but Robson insists he is going nowhere.
He told the club's official website: "Danny will be going nowhere. He's one of the lads I want to keep within the squad and there's no chance of him going anywhere."
