The 41-year-old, who scored 94 goals for the club during a six-year spell as a player, replaces Gary Caldwell and takes up his first role in management, having left his position as Nottingham Forest's under-23's boss.
Lester told the club's official website: "I am really proud to have been given this opportunity and I'm aiming to build something special."
Caldwell, who oversaw relegation last season, was sacked after a poor start to the League Two season which saw the Spireites lose five of their first eight games.
They lost both games under caretaker Guy Branston, who left the club on Thursday.
Director Ashley Carson added: "I spoke to a number of people who expressed interest in the job and it soon became clear that Jack was the outstanding candidate.
"Jack has impressed at the start of his coaching career as part of Nottingham Forest's highly respected academy set-up and he now feels ready to take on the challenge of a managerial role.
"He has the hunger and desire to succeed in management and he has set out his vision for the future of the club.
"As someone who already has the respect of the fans from his time as a player at the club, I am sure that Jack will receive all the backing he needs from the supporters as he embarks on his managerial career.
"Jack's return will hopefully herald the start of an upturn in the fortunes of the club."
Lester will be assisted by Nicky Eaden, who leaves his role as senior development coach at Coventry.
