Liverpool midfielder Jordan Henderson will captain England against Spain with Wayne Rooney set to start on the bench after missing training due to a minor injury.
Ryan Bertrand was also forced to sit out England training due to a knock ahead of Tuesday's friendly with the former World and European champions at Wembley.
The FA have also confirmed that 17-year-old Spurs midfielder Dylan Duncan has joined the training session with only 19 England players taking to the pitch at Tottenham's Enfield base.
"I wouldn't start Wayne in this game anyway. Jordan Henderson will be captain," Southgate told a news conference.
"The key for me is I believe we have several leaders and I don't think we can have just one captain.
"Too much responsibility has been on Wayne, we need to share that. Once you have a team of those leaders you can have a lot of success."
Manchester United captain Rooney was handed a recall for the 3-0 World Cup qualifying win over Scotland on Friday after starting as a substitute in the 0-0 draw in Slovenia last month.
England's interim manager Gareth Southgate is favourite to land the job on a permanent basis following the wins over Malta and Scotland and a draw in Slovenia helped England to the top of Group F with 10 points from four matches.
